AbstractFourier transform infrared spectroscopy was applied to the conformational analysis of somatostatin in H₂O, ²H₂O, dimethyl sulphoxide (DMSO)-²H₂O and DMSO. The results suggest that in these solvents the peptide adopts a similar structure. In agreement with circular dichroism (CD) spectroscopic data, evidence for turn structures was found; however, there was no evidence for the intramolecular β-sheet secondary structures proposed on the basis of Raman and CD spectroscopic results. Rather, the amide C=O and NH groups not involved in turns appear to be either hydrogen bonded to the solvent or free.
